氧化还原响应型连接臂增强鼠李糖修饰肿瘤疫苗活性的研究

摘要:肿瘤相关糖抗原sTn在肿瘤细胞表面大量异常表达,是开发肿瘤疫苗的重要靶点之一。传统疫苗构建策略是将糖抗原与载体蛋白偶联,然而载体蛋白具有较强的免疫反应,对sTn具有抑制作用。前期研究中发现免疫原性较弱的载体蛋白通过修饰鼠李糖等半抗原,能够招募内源性抗体并靶向抗原呈递细胞,增强糖抗原的免疫反应。本文在此基础上,设计了氧化还原响应型连接臂将鼠李糖修饰于载体蛋白,构建新型sTn肿瘤疫苗。体外实验和小鼠免疫评价发现,该氧化还原响应型连接臂能够促进溶酶体逃逸和抗原释放,有效增强sTn糖抗原的免疫反应,且所生产的抗体能够识别并杀伤肿瘤细胞。该策略有效提高了疫苗的免疫活性,有望广泛用于疫苗的构建。

Redox-responsive linker improves the immunogenicity of Rhamnose-modified anti-cancer vaccine
Abstract:Tumor-associated carbohydrate antigens, such as sTn antigen, which are abundantly exposed on the surface of cancer cells, are one of the most important targets for the development of anti-cancer vaccines. Traditionally, the carbohydrate antigens are conjugated with carrier proteins to form glycoconjugate vaccines. However, the carrier proteins usually elicit too strong immune responses that suppress the immune responses against carbohydrate antigens. Our previous studies found that the carrier protein modified with Rhamnose (Rha) hapten could significantly enhance the immune responses against sTn. The Rha hapten can recruit endogenous anti-Rha antibodies and then target antigen-presenting cells to improve the antigen delivery. In the present study, we constructed a novel sTn-based anti-cancer vaccine by using a redox-responsive disulfide bond linker to carry the antibody-recruiting hapten Rha. The in vitro study and mouse study revealed that this redox-responsive linker can facilitate the lysosome escape and accelerate the release of antigen following with dramatically increased sTn-specific immune responses. The induced antibodies can successfully recognize the cancer cells with sTn expression and mediate complement-dependent cytotoxicity against cancer cells. This strategy of vaccine construct efficiently improves the efficacy of vaccine and could be further applied in vaccine industry.
