RNA N4-cytidine acetylation in Arabidopsis thaliana
Abstract:RNA acetylation on N4-cytidine (ac4C) has recently been discovered as a novel modification on mRNA. The ac4C modification was shown to be a key regulator of RNA stability, RNA translation, and the thermal stress response. However, its existence on eukaryotic mRNA is still controversial. In plants, the existence, distribution pattern and potential function of ac4C are largely unknown. Here we report the presence of ac4C on the mRNA of both Arabidopsis thaliana . By comparing two ac4C sequencing methods, we found that acRIP-seq but not ac4C-seq was suitable for plant ac4C sequencing. We present transcriptome-wide atlases of ac4C in Arabidopsis thaliana mRNAs obtained by acRIP-seq. Analysis of ac4C distribution showed that the ac4C concentration in Arabidopsis mRNA was near the start and end sites of translation.
